Study of invertase from yeast isolated from sugar industry

Invertase from two yeast strains Candida humicolus 5M2 and Saccharomyces cerevisiae 7M3.2.1, isolated from Mitra Phuveing Sugar Industry, were studied. They were cultured in complete medium enriched with 10% sucrose by aerobically at 30 deg C at log phase and subjected to partial purification for extracellular and intracellular invertase. The purification was done by (NH4)2S04 precipitation and HPLC with IEC-DEAE ion-exchange column. Major fraction of extracellular invertase from Candida humicolus could be separated by 60-100% (NH4)2SO4 followed by DEAE column chromatography as a single active fraction at the concentration of 0.3-0.35 M NaCl with molecular mass of 110 kDa. The intracellular enzyme was partially separated by DEAE column chromatography at the same conditions. Two forms of intracellular invertase, high molecular weight form (200 kDa) and low molecular weight forms (58 and 64 kDa) were found in Saccharomyces cerevisiae. The high molecular weight form from crude cell extract from Saccharomyces cerevisiae was further separated by DEAE column chromatography at the concentration of 0.21-0.24 M NaCl.
