Microplate assay of glyoxalase I activity in blood samples

English. The activity of glyoxalase I enzyme has been proposed as a prospective diagnostic marker for the development of late complications of diabetes mellitus. The aim of the present study was to standardise the conditions for blood sample handling and storage in clinics, as well as to develop a simple spectrophotometric assay of glyoxalase I enzyme activity on 96-well UV plates. To test the influence of long term storage on glyoxalase I activity, the samples were kept at + 4 deg C, - 20 deg C, and - 80 deg C for up to four weeks. After that the blood cells were lysed and the supernatant was the used for the measurement of glyoxalase I activity. Our resulyts demonstrate that freezing at - 20 deg C of collected blood samples leads in glyoxalase I activity, which cannot be reversed by addition of protease inhibitors. We conclude that blood samples can be kept at + 4 deg C for up two weeks without any essential loos in enzyme activity and that - 80 deg C freezer is the most suitable choice for the long-term storage of samples species was: rat mouse human~= cow dog rabbit~= cat.

Latvian. Glioksalāze ir enzīms, kas organismā detoksificē daļu dziļās glikēšanas galaproduktu, tādējādi tās aktivitāte varētu noderēt par diagnostiku rādītāju diabēta vēlīno komplikāciju attīstībai. Pētījuma galamērķis bija standartizēt asins paraugu ņemšanu un uzglabāšanu apstākļos, kas būtu ērti izmantojami ikdienas klīniskajā praksē, kā arī izstrādāt metodiku glioksalāzes I aktivitātes spektrofotometriskai noteikšanai 96 lauciņu UV platītē. Lai izvērtētu paraugu glabāšanas apstākļu ietekmi uz glioksalāzes I aktivitāti, tie tika glabāt + 4 grādi pēc C, - 20 grādi pēc C un – 80 grādi pēc C , nosakot glioksalāzes I aktivitāti četru nedēļu laikā. Asins paraugi pēc uzglabāšanas tika lizēti, un GO-1 aktivitāti noteica pēc centrifugācijas iegūtajā supernatantā. Pētījuma rezultāti rāda nozīmīgu glioksalāzes I aktivitātes samazināšanos, asins paraugus uzglabājot - 20 grādi pēc C, turklāt aktivitāti neizdodas saglabāt, pievienojot dažādus proteāžu inhibitorus. Savukārt līdz pat divām nedēļām nekonstatē nozīmīgu glioksalāzes I aktivitātes samazināšanos asins paraugos, kas uzglabāti + 4 grādi pēc C. Tomēr ilgstošākai uzglabāšanai līdz vienam mēnesim rekomendējams izmantot paraugu sasaldēšanu - 80 grādi pēc C. Izmantojot izstrādāto metodi, noteiktā glioksalāzes I aktivitāte dažādu sugu indivīdu asins paraugos dilstošā secībā ir šāda: žurka pele cilvēks ~= govs suns trusis ~= kaķis.
