Mitochondrial hog-renal-cortex respiration system activity as a function of taken-out-organ storage conditions

The suitability of Collins solution - 4 mmol/l K2HPO4, 15 mmol/l KH2PO4, 16 mmol/l KCL, 10 mmol/l NaHCO3 (94 mosmol/l, pH 7.4) and its variant with novocain (0.1 g/l), used in kidney transplantation medicine was studied for storing hog kidneys appropriated for making mitochondrial preparations and studying respiration processes. After the set time of incubation - 2.24 and 48 hours, mitochondria were isolated from renal cortex and respiratory control ratio RCR and ADP/O ratio of obtained preparations were determined. It was revealed that hog kidneys taken-out from animals intended for slaughter may be protected against loosing their metabolic activity through incubation in cooled down Collins solution. If the incubation time will not exceed a few hours, the organs are suitable for making mitochondrial preparations and studying respiration processes. Worse condition of mitochodria obtained from organs incubated in Collins solution with novocain - used to protect the kidney intended for transplantation against angiostenosis - indicate that one can not go by a simple rule of 'selection by analogy' when selecting incubation solution. Desicion on using a new, or modified, medium should be preceded by analysis that excludes a negative effect of this components on the state of preparation obtained from incubated organ and/or tissue
