Suitability of BBL MGIT growth medium in veterinary mycobacteriology

A comparison has been made between the BBL MGIT growth medium (Becton Dickinson) with 4 conventional solid media (Middlebrook 7H10, Stonebrink, LJ medium with pyruvate and LJ medium with glycerol). Organ specimens obtained from 929 animals were decontaminated with N-acetyl-L-cysteine (NALC)-NaOH using the standard method. Mycobacteria were isolated from 391 of those specimens. The mycobacteria were recovered on MGIT from 323 specimens (82.61 %), on LJ-P from 266 specimens (68.03 %), on Middlebrook 7H10 from 222 (56.78 %), on LJ-G from 251 (64.19 %) and on Stonebrink from 231 specimens (59.08 %). MGIT recovered 40 strains, while only 8 strains were recovered on LJ-P, 6 strains on LJ-G and on Stonebrink and 3 strains on Middlebrook 7H10. The mean detection time of mycobacteria in MGIT was 15.26 (2 - 68) days, on Stonebrink it was 25.33 (5 - 56) days, on Middlebrook 7H10 25.78 (7 - 63) days, on LJ-P 26.56 (5 - 68) days and on LJ-G 30.33 (5 - 64) days. Using MGIT reduced the detection time by 10 days on average and by not using MGIT the results from 40 specimens (4.31 %) would have been false negative.
