Biodiversity and distribution of phytoplasmas associated with yellow leaf syndrome (YLS) in Cuba | Biodiversidad y distribución de los fitoplasmas asociados con el síndrome del amarillamiento foliar (YLS) en Cuba

English. Recent studies have confirmed the widespread of Yellow Leaf Syndrome (YLS) throughout Cuba, being the central and western regions the most affected areas. Sugarcane yellow leaf phytoplasma (SCYLP) was found in 99.3% of symptom plants; however, isolates have not been characterized yet. Polymerase chain reaction (PCR) products from 381 leaf samples with and without YLS symptoms, collected in previous surveys in 2002, and containing SCYLP phytoplasma, were digested with RsaI and AluI restriction enzymes. Polymorfic samples were selected and total DNA used as template to amplify the intergenic 16/23S region by a nested PCR (nPCR) assay with specific primers P1, P4, P7. P4/P7 PCR products were purified and used for direct sequencing. Intergenic sequences obtained were compared with those from 22 GenBank reference phytoplasmas, including those associated with Aster Yellows (16SrI) group previously reported in 1999. Phylogeny revealed a wide biodiversity related to YLS phytoplasmas present in the country since phytoplasmas from 16SrI, 16SrIII (X-disease) groups and a possible new 16SrI subgroup were identified. Results indicate that the main phytoplasma circulating in Cuba belongs to a new 16SrI subgroup called Cuban Sugarcane Yellow Leaf Phytoplasma, CScYLP (Cuba).

Spanish; Castilian. Estudios recientes han confirmado la diseminación del Síndrome del Amarilleamiento Foliar (YLS) por toda Cuba, donde el centro y oriente fueron las regiones más afectadas. El fitoplasma causante del amarilleamiento foliar de la caña de azúcar (SCYLP) fue encontrado en el 99.3% de las plantas con síntomas, sin embargo, aún los aislados no han sido caracterizados. Los productos de la reacción en cadena de la polimerasa (PCR), de 381 muestras foliares con y sin síntomas de YLS, colectadas en muestreos previos en 2002, y que contenían al SCYLP, se digirieron con las enzimas de restricción RsaI y AluI. Se seleccionaron las muestras polimórficas y su ADN total se utilizó como molde para amplificar la región intergénica 16/23S mediante un ensayo de PCR anidada con los iniciadores P1, P4 y P7. Los productos de PCR P4/P7 fueron purificados y empleados para la secuenciación directa. Las secuencias intergénicas obtenidas fueron comparadas con las de 22 fitoplasmas de referencia del Banco de Datos, incluyendo las asociadas al grupo del Amarilleamiento del áster (16SrI), informadas en 1999. La filogenia reveló una amplia biodiversidad relacionada con los fitoplasmas asociados al YLS, presentes en el país a partir de que se identificaron los grupos 16SrI, 16SrIII (Enfermedad X) y un posible nuevo subgrupo 16SrI. Los resultados indican que el principal grupo fitoplásmico circulante en Cuba pertenece a un nuevo subgrupo denominado Fitoplasma Cubano del Amarilleamiento Foliar, CScYLP (Cuba).
