Isolation and identification of indigenous Azospirillum species in Golestan province soils

Azospirillum species in association with wheat roots are able to supply the plant with part of its nitrogen requirement as well as produce plant hormones and sidrophores which cause improved root development and result in better moisture and nutrient absorption by the healthier roots. To evaluate the effects of Golestan indigenous Azospirilla on wheat (Triticum asetivum L.) yields, 150 soil and plant samples were collected from Golestan provinces. One hundred and twenty two isolates capable of forming an veil-like pellicle in a semi solid medium, free of N and with Brome Thymole Blue (NFB), were subjected to primary isolation and characterization .Subsequently the collection will be classified into the different species following purification on Congo Red Agar medium (CRA), NFB solid medium and biochemical tests. Fifty six isolates were capable formed colony dark red on CRA. Twenty four isolates were capable formed colony pinkish on Potatoes Agar (PA), white and small colony on NFB solid medium. Twenty four isolates were tested and classified into 3 species, based on their growth in a 3% medium of sodium chloride, biotin requirements, consumption and production of acid from glucose and production of polymorphous cells in a NFB medium. Among the identified species, A.brasilense was the highest in number, flowed by A.lipoferum and A.irakense.
