Method for increasing the efficiency in finding productive crosses in maize

Innovative methods in reducing the cost while increasing the efficiency of breeding work are of great help to maize breeders. This study aimed to evaluate the usefulness of cluster-based grouping using SSR and morphological markers in finding productive crosses (PC) in maize. In this study, the authors used four datasets, involving 192 crosses, to demonstrate the usefulness of this approach. The inbred-dialled crosses comprised of two sets, namely: set A- 55 crosses (yellow,), Set B- 36 (white). The inbred design 2 (Set C) consisted of 56 crosses (yellow). The non-inbred diallel (Set D) composed of 45 crosses (white). To evaluate the yield performance, the four sets of crosses including the check varieties were laid out in an alpha lattice design with three replications. For Set A, B, and C, the authors used respectively, 39,40, and 30 informative SSR markers to analyze the genetic relationship and grouping of the inbred lines. The DNA extraction, PCR, silver, straining, and band scoring were performed using protocols of CIMMYT-AMBIONET. Only markers with 20% heterozygosity and lines with 15% missing data were included in the analysis. For the Set D, the authors used 19 agromorphological traits in grouping with parent varieties. Using the four data sets, the author observed that inter-group crosses had greater number of PCs than those in intra-group crosses. In the Set A, 64% of the PCs came from the inter-group crosses in Set B,C and D. Further, SSR and morphology-based grouping followed by factorial crosses appeared more efficient by 12% than the conventional diallel crosses in identifying superior potential crosses in maize, thereby reduces the cost extensive field crosses and tests.
