Factors indicating cellular status during the cultivation of Spirulina platensis (Cyanobacteria)

To determine the most advantageous harvesting time in biomass production during the cultivation of Spirulina platensis, factors indicating cellular status such as dry cell weight, chlorophyll-a, pH in medium, coil number, floating activity and ultrastructure were monitored. As a whole, the change of examined factors followed a normal growth pattern, they steadily increased to the maximum in the stationary phase, and thereafter decreased. Partial exceptional patterns were found in pH, coil number and floating activity, showing that the former two factors sharply increased, and the latter decreased in the early culture period. The floating activity of senescent cells after the stationary growth phase was sustained over 79% within one hour, which resulted from the increase dimension of gas vacuoles rather than an increase in their number. The culture grown under the condition of a fixed pH of 9.5 was found to be 3-3.5 times higher in OD than that of the uncontrolled, suggesting that the pH control of culture medium could significantly contribute to the improvement of biomass production. Consequently, it can be concluded that the consensual changing pattern in the above-mentioned factors could be used as a criterion for determining harvesting time. Among the factors, the pH and floating activity could be readily used as a simple indicator of the determination of the harvesting time of the cells.
