Aspergillus terreus ASKU 10 was selected as the strain that produced the highest beta-glucanase in solid-state fermentation using rice straw as substrate. Factors, addition of rice bran, cultivation time, spore inoculums, initial moisture content, corn steep liquor (CSL) and KH2PO4 were optimized using "one-factor-at-a-time" method for beta-glucanase production. The highest beta-glucanase activity achieved under optimal condition as follows: no addition of rice bran into the 10 g rice straw, with 10**(7) spore inoculums, 5 days of cultivation time, 75 percent initial moisture content, 3.3 ml CSL and 0.825 g KH2PO4. Under this condition Beta-glucanase production with 4054 U/g dry substrate has been obtained.
