Chinese. 研究POU1F1基因第6外显子HinfⅠ、AluⅠ和PstⅠ酶切位点多态性对郏县红牛生长发育性状的遗传效应。以144头郏县红牛为研究材料,用PCR-RFLP方法检测POU1F1基因第6外显子HinfⅠ、AluⅠ和PstⅠ基因座多态性,并分析其基因型间遗传变异与生长发育的关系。451 bp的POU1F1基因第6外显子片段被HinfⅠ酶切后表现多态;AA、AB和BB基因型频率分别为0.187 5,0.305 6和0.506 9,A和B等位基因频率分别为0.340 3和0.659 7,处于Hardy-Weinberg不平衡状态;该位点纯合度为0.551 0,杂合度为0.449 0,有效等位基因数为1.814 8,多态信息含量为0.348 2,属中度多态,可作为该群体遗传资源评价的建议性指标。而POU1F1基因第6外显子AluⅠ和PstⅠ基因座均未检测到多态性,仅发现AA基因型,表明这些基因座比较保守。HinfⅠ多态性与郏县红牛生长发育性状的相关分析表明,不同基因型与郏县红牛体高、腰角宽、胸围、尻长、十字部高、管围、体质量等指标均无显著相关性(P0.05); BB基因型个体体长显著高于AB和AA基因型个体(P0.05),提示B等位基因可能对体长有显著影响。BB基因型可作为郏县红牛体长性状标记辅助选择的DNA标记。

The objective of the present study was to study the genetic effects of polymorphisms of HinfⅠ, AluⅠand PstⅠloci within POU1F1 gene exon 6 on Jianxian Red cattle growth traits. POU1F1 gene exon 6 polymorphic fragments were detected by PCR-RFLP technique using 144 Jianxian Red cattle. Association analysis of POU1F1gene exon 6 polymorphism with growth traits was also conducted. The results showed that 451 bp PCR products digested with HinfⅠdemonstrated polymorphisms. Moreover, the frequencies of allele AA, AB and BB genotype of Jiaxian red cattle were 0.187 5, 0.305 6 and 0.506 9, accordingly, the frequencies of allele A and B were 0.340 3 and 0.659 7 respectively. The homozygositiy, heterozytosiy, the effective allele numbers, and PIC value of POU1F1 gene exon 6 HinfⅠlocus were 0.551 0, 0.449 0, 1.814 8 and 0.348 2 respectively. As this locus belonged to the middle polymorphic index, it was suggested to be an effective evaluation parameter for studying genetic resource. However, no polymorphism was found at POU1F1 gene exon 6 AluⅠand Pst I loci. The correlation of HinfⅠpolymorphisms of POU1F1 gene exon 6 and growth traits was analyzed and the result showed that there was significant relationship between Hinf I polymorphism and body length (P0.05). The individual with genotype BB had longer body length than the individual with genotype AB and AA, suggesting the allele B had significant effect on body length of Jianxian red cattle. The genotype BB was significantly correlated with the fine performance and the selection of the allele B was better for the improvement.
