One feature of different stages of gut problems is the disruption of tight junctions, leading to a loss of integrity across the intestinal barrier. In this experiment, Caco-2 human adenocarcinoma cell lines were used as a model for the intestinal epithelia. Transepithelial electrical resistance (TEER) measurements indicate Caco-2 monolayer integrity. pH-controlled faecal batch continuous culture fermentations were used to study in vitro the distribution of the main micro-organisms involved in tryptophan metabolism. Twelve hours of exposure to tryptophan fermentation products was enough to modify TEER measurements. The fermentation products were different depending on the microbiota involved in the amino acid fermentation. Escherichi coli and Citrobacter freundii were present after 13 days of fermentation and the combination of both decreased TEER and open up the spaces between confluent cell cultures. However, our results indicate that the response of the cells tight junctions varied according to the biodiversity of microorganism present at the end of the experiment in the batch cultures.
