Preparation of specific Rose Bangal plate Test Antigen for Diagnosis of Sheep Brucellosis

Rose bengal antigens were prepared using Brucella abortus 19 or 99 and Brucella melitensis Rev. 1 strains. The final concentrations of 5%, 6% and 8% of bacterial germs and PH of 3.6+0.05 were adjusted. 3 batches of antigens were prepared from each strains (Totally 6 batches). The survey was carried down in 2 groups of animals. The first group, 100 sheep, 80 peregnant and vaccinated, (20 as control infected experimentally with virulent strain), were bled and their sera (a total of 1000 sera) were tested with the prepared antigens and other serological tests imultaneously. The results were compared with those sera of the second group (1000 sera) which were collected from naturally infected flocks all over the country sent by the veterinary organisation. The final results were summarised and evaluated and it was concluded that: 105% brucella strains 19 or 99 antigens showed higher sensitivity. 2- 8% brucella 19 or 99 antigens showed a higher specifity and it is suggested that 8% antigens strains 19 or 99 brucella abortus should be used in test and slaughter of sheep brucellosis control and 5% antigens strains 19 or 99 should be used in the brucellosis control screening program.
