Operating conditions and availability of the swab method for the quantitative assay of proteins attached onto stainless steel

Swabbing conditions were studied for the quantitative assay of various amounts of bovine serum albumin (BSA) or egg albumin (EA) attached onto stainless steel. We used a swab stick comprising polyethylene (PE) head and polypropylene (PP) shaft that was non-reactive to reagents of the Lowry-Folin assay used in this study. Swabbing was conducted at 80 to 140 mN for 20 s using 0.1 M NaOH as a wetting solution for the PE-head swab. The collected proteins on the PE-head swab were eluted efficiently in 0.1 M NaOH containing 0.1% sodium dodecyl sulfate as an eluent. As a result, the proteins of at least 1 to 700 microg could be quantified with the recovery efficiency of 97%. The wetting solution was found to be essential to recover the proteins from stainless steel. It was confirmed that EA was detected at a recovery efficiency of more than 92% even though EA was preheated at 40 to 120degC, beyond which the recovery efficiency decreased markedly. These results showed the fundamentals of the operating conditions for the high performance of the swab method and its availability.
