Study of simple methods to estimate number of muscle fibers by using M. biventer cervicis of chicken

In the present experiment we used the planimeter method and a method using paper weight to estimate the number of muscle fibers in M. biventer cervicis cross section of chicken. Actual counts of the number of fibers were also made to investigate which of the estimations more closely approached the actual values. M. biventer cervicis was obtained from 6 male chickens (White Leghorn, commercial). Frozen sections were prepared with a cryostat and then stained with NADH-tetrazolium reductase (TR) to make test specimens. Fractional photographs of the cross sections with the largest cross-sectional areas were taken under a microscope at a magnification of x20, and those photos were then combined to reproduce a photo of the entire cross section at a maximum magnification of x105. Using these reproduced photos, actual counts of the number of muscle fibers and estimates using the planimeter and paper weight methods were made. The degree of error between the actual numbers and each of the estimated values was calculated. We found that estimates using both the planimeter and paper weight methods closely approximated the actual counts. However, we found that in the degree of error of the estimates against the actual count values, the arcsine conversion value tended to be smaller with the paper weight method than with the planimeter method, and moreover that the degree of error itself was significantly smaller (p0.05). Therefore, the paper weight method is considered more suitable than the planimeter method in estimating numbers of muscle fibers.
