A Simple Preparation of Multi-channel Microelectrode Array

This paper provides a detailed protocol for making a relatively simple multi-channel array of tungsten microelectrodes, spaced by the thickness of tubing, to record multi-unit activity in the visual cortex. The technique involves (1) inserting a microelectrode inside a tube, (2) arranging the microelectrode and tube assemblage into a layer, (3) soldering the end of each microelectrode to a connector, and (4) stacking several layers together to make an array. The array can be constructed easily in most laboratories and manipulated much like a single microelectrode, and thus can be used for recording in any cerebral cortex. Two types of arrays (a straight or bent type) were used. Success rates of about 35% per microelectrode were obtained, with some microelectrodes picking up activity from more than one cell. The number of microelectrodes can be increased to more than one hundred by adding more microelectrodes in parallel. The array can be advanced in the same location of the cortex, without any depression of the cortex. Variations in the dimensions in the vertical or horizontal axis may be possible by adjusting the length from the tip of microelectrode to the tubing, or altering the thickness of tubing.
