Quantitative trait loci (QTL) for P uptake have been previously identified in a mappping population derived from across between Nipponbare (spp. japonica, intolerant) and Kasalath (spp. indica/aus, tolerant) and among these was a major QTL, Pup1. The Pup1 locus was recently fine mapped on rice Chromosome 12 and the corresponding 272 kb genomics region was sequenced in the tolerant donor landrace Kasalath. P-deficiency tolerant rice varieties are now being developed using highly specific gene-based PCR markers that were designed based on the Pup1 sequence. These markers are used to (1) assess the haplotype of Pup1 in diverse rice germplasm; (2) identify recipient parents for Pup1 introgression; and (3) select for Pup1 in the breeding process. The development of Pup1 introgression lines is ongoing at IRRI using modern irrigated indica varieties (IR64, IR74) and an upland variety (Ashoka 2228) as recipients to assess if Pup1 is beneficial in diverse genetic backgrounds and environments. First field screening will b conducted.
