Regeneration frequency from in vitro derived calli of rice (Oryza sativa L.)

The studies on the induction and maintenance of callogenesis and achieving high frequency regeneration of potential plantlets of different rice varieties through tissue culture technique in vitro were performed. For callus induction 2,4-D at the concentrations of 2, 3 and 5 mg/l, for shoot initiation and its elongation, BAP at the concentrations of 2, 2.5 and 3.0 mg/l and for root induction IAA and its elongation NAA was used at the concentrations of 1.0, 1.5 and 2.0 mg/l. Five commercial rice varieties (Khushboo, Sarshar, Shua-92, Mahak and Shadab) were examined for callus induction regeneration. The MS media containing 2 mg/l 2,4-D took significantly lesser number of days (4.20) to callus induction and increasing concentration delayed callus induction. MS media + 2,4-D at 3 mg/l resulted callus induction in 4.73 days and at 2 mg/l callus induction was recorded in 4.93 days.
