Molecular Analysis of the Goat Lipoprotein Lipase and Acetyl-Coenzyme A Carboxylase Alpha Genes

Using cDNA from 18 goats belonging to three breeds, we have characterized the complete coding sequence and the most part of it of the LPL and ACACA genes, respectively. Two single nucleotide polymorphisms (SNP) were detected in the goat LPL cDNA sequence: a G50C missense mutation, and a T2094C substitution in the 3'UTR. One silent SNP was identified at exon 45 (C5493T) of the ACACA cDNA sequence. These polymorphisms are segregating at variable frequencies in five analysed breeds. The LPL G50C SNP was associated with fat yield (P equal to 0.04) and tended to affect the milk dry weight basis (P equal to 0.07), however, the T2094C SNP was not associated with any of the measured traits. The ACACA mutation is associated significantly with fat yield (P equal to 0.02), lactose content (P equal to 0.01) and somatic cell content (P equal to 0.03).
