The investigation of Heat Shock Protein (HSP 27), Malondialdehyde (MDA), Nitric Oxide (NO) and Interleukin (IL-6, IL-10) levels in cattle with Anaplasmosis | Anaplasmosisli sığırlarda Isı Şok Protein (HSP), Malondialdehit (MDA), Nitrik Oksit (NO) ve İnterlökin (IL-6, IL-10) düzeylerinin araştırılması

English. Blood parasitic diseases such as tropical theileriosis in cattles are observed in Turkey. On the other hand, babesiosis and anaplasmaosis are other primer tick-borne diseases. Anaplasmosis is generalised in all over the region of Turkey. In the present study, we aimed to reveal the alterations of immune system and oxidative stress in cattles with anaplasmosis by analysing heat shock protein (HSP 27), malondialdehyde (MDA), nitric oxide (NO), interleukin-6 (IL-6) and interleukin-10 (IL-10) levels. The research has been performed on 15 healthy and 15 cattles with anaplasmosis, aged 1-3 years old. The plasma levels of malondialdehyde and nitric oxide were determined by spectrophotometric methods while the concentrations of heat shock protein and interleukin (IL-6 and IL-10) were detected by using commercial ELISA kits. The levels of plasma MDA (P 0.001), NO (P 0.001), HSP 27 (P 0.01) and serum IL-6 levels were determined significantly higher in cattles with anaplasmosis than the control group. The levels of serum IL­ 10 was not showed significant differences between the control and the patient group. The increases in the levels of malondialdehyde, nitric oxide, HSP 27 and interleukin-6 of the cattles with anaplasmosis may indicate the stimulation of host immune system and the existence of oxidative stress.In conclusion, determination of the levels of malondialdehyde, nitric oxide, HSP 27 and interleukin-6 might be a indicator for diagnosis of cattle anaplasmosis.

Kan parazit hastalıkları sığırlarda sıklıkla görülmekte olup, Türkiye'de de oldukça yaygındır.Bu hastalıklardan tropikal theileriosis en yaygın görülen kan paraziti hastalığıdır.Babesiosis ve anaplasmosis ise diğer önemli kan paraziti hastalıklarındandır. Anaplasmosis Türkiye'de her bölgede görülmektedir. Bu çalışmada anaplasmosisli sığırlarda oksidatif stres ve immun sistem değişimlerinin ısı şok protein (HSP 27), malondialdehit (MDA), nitrik oksit (NO), interlökin-6 (IL-6) ve interlökin-10 (IL-10) parametreleri ile ortaya konulması amaçlanmaktadır. Çalışma, 1-3 yaş arasında 15 sağlıklı ve 15 anaplasmosisli sığırda gerçekleştirilmiştir. Çalışmada malondialdehit ve nitrik oksit düzeyleri spektrofotometrik yöntemler ile, HSP 27 ve interlökin (IL-6 ve IL-10) düzeyleri ise ticari ELİSA kitleri kullanılarak belirlenmiştir. Anaplasmosisli sığırlarda plazma malondialdehit (P 0.001), nitrik oksit (P 0.01), HSP 27 (P 0.01) ve serum interlökin-6 (P 0.001) düzeyleri kontrol grubuna göre yüksek bulunmuştur. Serum interlökin-10 aktivitesinde ise, kontrol grubu ile anaplasmosisli sığırlar arasında önemli bir fark bulunamamıştır. Anaplasmosisli sığırlarda MDA, NO, HSP 27 ve IL-6 düzeylerindeki artış, anaplasma etkeni tarafından konak savunma sisteminin uyarıldığının ve bu esnada oluşan yangı ile birlikte oksidatif stresin meydana geldiğinin göstergesi olabilir. Elde edilen sonuçlara göre, anaplasmosiste malondialdehit, nitrik oksit, HSP 27 ve interlökin-6 düzeylerinin belirlenmesi, hastalığın tanısında faydalı olabileceği kanısına varılmıştır.
