Immobilized protoplasts as alternative sources of pharmacologically important secondary metabolites production are analyzed on the example of Catharanthus roseus plants. The yield of leaves' mesophyll protoplasts in dependence on cellulase concentration, time of incubation and temperature of isolation medium is studied. The impact of cellulase concentration, incubation time and medium temperature is calculated for 3 and 16 hours, 1% and 2% cellulase concentration and average temperature of the experiment equals 26 deg C. The influence of cultivation conditions on the immobilized protoplasts physiological status (increase of biomass and chlorophyll content) is evaluated for the period of 7 days. Total alkaloid content in immobilized protoplasts is determined in dependence on time continuance and incubation regimes. According to the experiment it is possible to determine the conditions of direct stimulation and excretion of indole alkaloid in immobilized protoplasts of Catharanthus roseus plants.
