[目的] 建立薄层色谱―蒽酮比色法定量测定低聚乳果糖含量的方法。[方法] 分别以蔗糖、葡萄糖、乳糖、可溶性淀粉为碳源, 酵母膏、蛋白胨、牛肉膏、尿素为有机氮源, 不同硫酸盐为金属离子添加形式, 在节杆菌产生的果糖甘酶的作用下合成低聚乳果糖样品, 用薄层色谱―蒽酮比色法定量测定样品液中低聚乳果糖的含量。[结果] 薄层层析展开剂为正丙醇∶水= 9∶1 时, 样品展开效果较好; 根据低聚乳果糖标准液的吸光度值建立的标准曲线在1 .781 3 ~17 .812 5 mg/ ml 范围内具有良好的线性关系( R2 =0 .993 6) ; 样品液在蒽酮显色后2 h 内的吸光度值变化较小, 性质较稳定。[结论] 薄层色谱―蒽酮比色法适合用于低聚乳果糖含量的定量测定。
Show more [+] Less [-]English. [Objective] The study was to establish the method of quantitative determination on lactosucrose content by thin-layer chromatography-anthracenone colorimetry . [Method] With sucrose , glucose , lactose , soluble starch as carbon sources , yeast extract , peptone , beef extract , urearesp as organic organic nitrogen source and different sulfates as additive types of metal ions , the lactosucrose samples were synthesized under effect of glucosidase which produced by arthrobacter . The lactosucrose content in samples was determined by chromatography ―anthracenone colori metry . [ Result] When the developing solvents of thin-layer chromatography was n-propanol ∶water = 9∶1 , the developing effect of the sample was better . The standard curve established according to the absorbance value of sample solution had a good linear relationship in 1 .781 3 ~17 .812 5 mg/ ml( R2 = 0 .993 6) . The absorbance value of the sample changed little and its property was stable in 2 h after colouration with anthrone . [ Conclusion] The thin-layer chromatography ―anthracenone colorimetry was suitable for determining lactosucrose content .
