Chinese. 研究了NC89烟草悬浮细胞的最佳培养方案,为进一步提高辅酶Q10的产量提供理论依据。通过单因素试验和正交试验考察了培养基种类、蔗糖浓度、接种量、转速、pH值和装液量对烟草细胞生长及辅酶Q10合成的影响。结果表明:接种量对细胞的生长及辅酶Q10合成的影响作用极显著;蔗糖浓度对二者的影响均不显著,优选蔗糖浓度为20g・L. 1;装液量对辅酶Q10合成的作用显著,而对细胞生长作用不显著;优选转速为110r・min. 1,pH值为6.2。可见,适于NC89烟草细胞生长及辅酶Q10合成的优选培养条件为White基本培养基,附加蔗糖20g・L. 1,装液量为20mL(100mL三角瓶).接种量75g・L. 1,初始pH值为6.2.摇床转速为110r・min. 1。在该条件下细胞产量和辅酶Q10含量分别为16.653g・L. 1和14.780mg・L. 1。[著者文摘]

The optimal cultural conditions of tobacco cell NC89 were selected in order to provide a theoretical basis for further enhance the production of Coenzyme Q10 Factors as media types, sucrose concentrations, inoculation, rotational speeds, pH value and liquid volume which affected tobacco cell growth and CoQ10 synthesis were studied through single factor and orthogonal design. The results showed that cell growth and CoQ10 synthesis were remarkably affected by inoculation, the optimal concentration of sucrose was 20g・L. 1, CoQ10 synthesis was affected by culture volume, 110r・min. 1 and pH 6.2 were the prior choices respectively. The optimal culture conditions for NC89 cell growth and CoQ10 synthesis was White medium, concentration of sucrose 20g・L. 1, culture volume 20mL (100mL flask), inoculation 75g・L. 1, pH 6.2 and rotational speed ll0r・min. 1. And the cell biomass and CoQ10 content could reach 16.653g・L. 1 and 14.780mg・L. 1 respectively.
