Chinese. 研究嗜水气单胞菌的大量培养和灭活方法,为制备嗜水气单胞菌疫苗提供切实可行的方法。选取毒力较强菌株,按照培养温度、振荡与否、培养基的pH值、加入的营养源等不同条件培养菌液,分别检测其溶血价及OD值;再以不同温度、时间、甲醛浓度制成疫苗,检验其残菌数、溶血价及抗原性。结果表明,以pH为7.5、φ=1%胰蛋白胨肉汤在28℃、180r/min振荡培养的菌液最佳;以φ=0.2%甲醛在28℃恒温培养箱灭活12h的疫苗抗原性最强,无菌检验和安全性检验合格。[著者文摘]
Show more [+] Less [-]English. The method for mass culture and inactivation of Aeromonas hydrophila was studied in order to provide feasible ways for generate vaccines. The test selected a highly virulent strain and preparated bacterial liquid from different culture conditions according to the cultural temperature, vibration, pH of broth nutrient source and respectively detected hemolytic titer and OD value;then preparated vaccine with different inactiva- ting methods according to different inactivating temperature, time, formaldehyde concentration and tested the remnant microorganism number, hemolytic titer and antigenicity. The results showed that the optimal cultural condition is pH 7.5,1% tryptone broth, 28 ℃, 180 r/min, and the vaccine inactivated with 0.2% formaldehyde at 28 ℃ for 12 h has the strongest antigenicity, and is qualified by sterility test and safety inspection.
