Chinese. [目的]探讨真姬菇单核体间RAPD标记多态性,为育种材料的选择提供一定的分子水平依据。[方法]以真姬菇(Hypsizygus marmoreus)工厂化栽培白色菌株SIEF3136的原生质体单核体和不同孢子单核体为材料,进行RAPD分析。[结果]筛选到38条多态性引物,共扩增341条条带,其中253条表现多态性,多态率为74.2%。31个菌株的相似系数为0.34~0.90。除单核体S18-16,同一交配型的单核体间的相似系数都在0.60以上,表现出较高的相似性。在大约0.61的相似水平上,所有菌株分为2组。PM-01与T1、T3聚为第1大类,PM-02与T2、T4聚为第2大类。另外,在不到0.62的水平上S18-16聚为第3大类,显示出极大的差异,这很可能是该单核发生突变所致。[结论]真姬菇子实体上担子在减数分裂中染色体交换很高,RAPD标记可以用于真姬菇单核多态性研究,并可为交配型鉴定提供参考。

[Objective]The research aimed to compare the extraction method of total RNA from _Pinus tabulaeformis _Carr,and to obtain high quality total RNA of _P. tabulaeformis _Carr mature embryo.[Method]SDS mehtod,CTAB method,Trizol method and improved SDS mehtod were employed to isolate total RNA from _P. tabulaeformis _Carr mature embryo.And the qaulity of RNA was examined with OD _value and agarose gel electrophoresis.[Result]The purity of RNA extracted was high by improved SDS mehtod,_A__260_/A__280_ _was 1.97 and _A__260_/A___230_ was 2.08.The bands of 28 S,18 S and 5 S were clear and the brightness of 28 S bands was twice of the bands of 18 S.After RT_PCR,ds cDNA bands size was range from 0.3 to 3.0 kb,the results further demonstrated that the purity and integrality of total RNA isolated using improved SDS method were significantly satisfactory for the demands of molecular biological research.[Conclusion]Improved SDS mehtod had advantages of low_cost,simple_operation,short_time,good quality of extracted RNA,and it was an effective method of the extraction of total RNA of polysaccharide and polyphenol plants._
