The aim of this work was to analyze the effects of forage type (alfalfa and grass hay) and time after feeding on the bacterial communities in ruminal solid digesta from sheep. In order to study bacterial diversity, DNA was isolated from samples of solid digesta sampled 0, 4 and 8 hours after feeding, and the ITS1 region of rDNA operon was amplified by PCR and analyzed by ARISA. A total of 109 peaks were detected in the ARISA electropherograms across the full set of 24 samples. Bacterial diversity, as assessed by Shannon index, was greater (P<0,001) for AL compared with GR diets. For both diets, bacterial diversity at 0 and 8 hours after feeding was greater (P<0.05) than that at 4 h post-feeding.
