Mycoplasmosis is an important disease of poultry worldwide and there is in all parts of the world and it has been reported from Iranian poultry house. This disease would be seen by itself or complex with other diseases. Pathogenic mycoplasma in poultry include Mycoplasma gallisepticum (Chronic Respiratory Disease) and Mycoplasma synovie (Infectious Synovitis),and these microbe not only increase mortality and economical loss, but also predisposing chickens to other diseases. Many antibiotics were used to control and reduce severity of the sickness, but they cannot eradicate this disease and the infected chickens has a career role. Isolation and determination of pathogenic agents and antibiotic sensitive evaluation with other controlling manner can reduce the economical loss of this disease. In this research sampling of slaughterhouse in Tehran province was done. The samples were investigated by serological tests like as Rapid Serum Agglutination (RSA), ELISA, Broth culture and Mycoplasma specified agar. The main test for isolation and determination in this survey was RSA. In this investigation 4530 pharyngeal swab and blood sample from 226 poultry house were taken, in RSA method with MG antigen 11.27% were positive and false positive. This evaluation for MS was 38.19%. After this step the ELISA test was done on the positive and false positive RSA results, and 66.86% of MG and 70.13% of MS were positive. The results indicated that in RSA method, false positive was high and specificity was low. Comparison between prevalence of MG and MS in different seasons revealed in winter there are the most prevalence (P0.001). In this research were detected mycoplasmas from 82 of 226 tested poultry houses by culture. The results revealed "MG 12", "MS 29", "MG and MS 5" and "non MG and MS 36". Keywords: Myco
