Cytochrome P450-mediated pyrethroid resistance in the diamondback moth, Plutella xylostella (L.)

In insects, cytochrome P450s are enzymes known for the catalysis of many different reactions involved in growth and development, tolerance to plant toxins and insecticide resistance. Insecticide resistance results from increased detoxification by cytochrome P450s via elevated transcriptional over expression. Although P450s have been implicated in the metabolism of insecticides in many insects, their role in the insecticide resistance of the diamondback moth Plutella xylostella has not been comprehensively explored, particularly at the molecular level. Understanding the molecular basis of insecticide resistance of the pest is often considered necessary for resistance management. To examine the overall contribution of an over expressed P450 CYP6BG1 to permethrin resistance of a laboratory selected permethrin-resistant strain of P.xylostella (ARS), the effects of silencing CYP6BG1 by RNA interference via double-stranded RNA (dsRNA) droplet-feeding on larval resistance and total P450 activities, were examined. Knockdown of CYP6BG1 transcript had corresponding reductions in resistance and total P450 activities of microsomal fractions of CYP6BG1 dsRNA fed ARS larvae, implicating an important role of CYP6BG1 in increased detoxification of permethrin. Knockdown of CYP6BG1 transcript in a field-derived permethrin-resistant strain (Kanoya) also showed concomitant decreases in resistance and total P450 activity of larvae fed CYP6BG1 dsRNA, supporting further the claim that CYP6BG1 is associated with resistance to permethrin. The data presented CYP6BG1 is associated with resistance to permethrin. The data presented here provide additional information on the molecular basis of insecticide resistance of the diamondback moth, which could facilitate development of tools for its effective management.
