Mullein (Verbascum thapsiforme Schrad.) is a native medicinal plant used for the treatment of inflammatory diseases, asthma, spasmodic coughs, diarrhea and pulmonary problems. This work discusses the extraction of mullein flower with different solvents, water and ethanol solution (five different concentration: 20%, 40%, 60%, 80% and 96%). Extractions were done in drug-solvent ratio of 1:20 (m/v). The content of total flavonoids and total phenols was determined by spectrophotometric method, using (+)-catechin (C) as standard for flavonoids (results were expressed as mg of catechin equivalent (CE) per g of dry extract - mg CE/g), i.e. using galic acid (GA) as standard for phenols (results were expressed as mg of gallic acid equivalent (GAE) per g of dry extract - mg GAE/ g). The highest values were achieved for mullein dry extracts obtained by 60% and 80% ethanol. Tests included identification and quantification of phenolic components with LC/MS/MS system on all dry extracts. The highest total amount of phenolic acids (1.0119 mg/g of dry extract) was determined in mullein extracts prepared with 80% ethanol. The antioxidative activity of dry extracts was tested by IC50 values using a standard DPPH method. The extracts obtained with 60% and 80% ethanol show the highest antioxidant effect.
