Evaluation and screening of Iranian rice germplasm with use of quantity and quality character and molecular marker

Study of genetic structure and variation in the plant germplasm is one of the most important components in plant breeding programs. In the present study, genetic diversity in 96 rice variety evaluated by using of 11 agro-botanical traits and 23 SSR Markers. The aim of the work was to study variation between varieties with molecular and morphological data and to use in a future breeding program. Extracted genomic DNA of each plant extended with 23 SSR primers and electrophoresed on denatured polyacril amid gel. Morphological traits was done in a field experiment in an augmented randomized complete block design with 5 replications. Totally 112 polymorphic bans were observed in the studied populations. Among the primers used, RM481 and RM592 showed the highest polymorphic band (10 bands) and RM443 and RM184 showed the lowest polymorphic band (2 bands). Cluster analysis of three populations showed two clusters, one local varieties and other is improved and foreign varieties. Cluster analysis of total varieties with UPGMA method by using of molecular data showed three groups of varieties (group 1 and 2 were local varieties group 3 was improved and foreign varieties). But cluster analysis of morphological data can not seprat local varieties from improved and foreign varieties. In conclusion SSR markers can be used as a power full tools for assessing inter and intra populations genetic diversity as well as genetic differentiation and identification.
