Quantitation of tyrosine-ortho-sulfate in human urine by ion-pair reverse-phase high-performance liquid chromatography.

We have earlier reported the development of a high-performance liquid chromatography technique for quantitative determination of free tyrosine-ortho-sulfate (TyrS) in human urine with a sensitivity up to 0.5pmol/analysis. Samples containing TyrS were chromatographed on a reverse-phase column using an ion-pair reagent containing tetrabutylammonium phosphate (TBA-P), and the TyrS eluted was determined based on the fluorescence displayed following post-column labeling with ortho-phthaldiadehyde. Using this procedure urine samples taken from fifty randomly selected healthy human subjects were analyzed, which revealed that the free TyrS level falls within the range of 6-88mug/ml with a mean value of 28.42+-16.39mug/ml. No sexual variation or age difference could be observed in the urinary tyrosine-ortho-sulfate content.
