Isoenzymes are widely used as genetic markers in biochemical, genetic, development, taxonomic and phylogenetic studies. Isozyme data can be used to characterize inbred lines, exotic hybrids and wild germoplasm in maize. Electrophoresis of isozymes provides an effective method for assessing purity in seed lots of maize during inbred maintenance and hybrid seed production. Many maize inbreds reveal a unique fingerprint pattern following the electrophoretic separation of seeding enzymes. Having first established the enzyme fingerprints of a hybrid and those of the constituent inbreds, it is possible to analyse hybrid and inbred seed lots in order to check for the occurrence of selfed seed in hybrid lots; outcrossed seed in inbred and hybrid lots; and otherwise rogue seed in inbreds and hybrid seed lots. Banding patterns for 8 enzymes systems (Acph, Adh, -Glu, Mdh, Idh, Pgm, Pgd, Phi) (15 loci) were carried out in inbred and hybrid coleoptil tissue extracts of maize in purity check of hybrid seed production. Being now able to determine seed quality, the information is transmitted to the producers and the marketing staff rapidly and efficiently.
