Polymerase chain reaction analysis of an intron in BmAntp gene [of Bombyx mori].

BmAntp gene of the domestic silkworm, Bombyx mori, is a homeotic gene related to the formation of thoracic segments. A part of genomic DNA and cDNA of the BmAntp gene has been cloned. However the intron region and 5' terminal of the gene were not isolated from genomic DNA. We attempted to measure the intron length of the gene by using the polymerase chain reaction (PCR). Four primers referenced from DNA sequences on the 3' and 5' flanking regions of the exon were prepared. In every reaction batch examined, PCR products were detected. Then, the PCR products were used for southern analysis to determine whether the amplified DNA originated from the BmAntp gene or not. Fluorogram of Southern analysis showed a different pattern from PCR electrophoregram. Consequently, the BmAntp gene was amplified by PCR.
