In vitro regeneration of mulberry (Morus indica L.) cv S54 has been achieved via direct organogenesis from mature leaf cultures. Explants when inoculated on medium supplemented with 2,3,5-triiodobenzoic acid (TIBA) did not show any morphogenic response. Thidiazuron (TDZ), a substituted phenyl urea with cytokinin activity, was significantly more effective in inducing nodule like structures from leaf cultures, which did not develop further. Regeneration was obtained from all the combinations of TDZ with indole acetic acid (IAA). However, the optimal TDZ and IAA concentration for organogenesis from leaf cultures was 4.54 microM and 1.71 microM, respectively. Regenerated shoots were rooted on Murashige and Skoog's (MS) medium containing 4.9 microM indole butyric acid (IBA) followed by transfer to the greenhouse.
