Phenotypic and genetic changes associated with embryo cultured Zea mays.

Embryo culture is now routinely used as an auxiliary tool in the conservation or generation of genetic variability. This study was conducted to see if embryo culture by itself can induce genetic changes. Embryo formation was initiated among different stocks of Zea mays (Lagkitan, Macapuno, Purple 3, Purple 4 and IPB Var 1). Embryos from 10, 12, 14, 16, 18 and 20 days post-pollinated ears were aseptically removed and inoculated in Murashigue and Skoog's medium supplemented with 5% coconut water with the scutellar side in contact with the medium. Root tips were collected during potting-out for cytological studies. Differences in plant vigor and plantlet differentiation (e.g. albino, curling of leaves, and callusing) within the same embryo age and among different embryo ages were evident. Abnormalities as to the development of floral structures such as apical cobs, tassel seeds and twin seedling were also observed. Variabilities in chromosome number were observed in root tip cells of the embryo-derived plantlets. Hyperbloids complemented hypoloids in frequency of occurrence. The significance of these findings in routine use of embryo rescue was discussed.
