Factors influencing the spontaneous contraction of the oviduct of Locusta migratoria in vitro.

The influence of some physiological and chemical factors on the spontaneous contraction of the isolated oviduct of Locusta migratoria migratorioides was examined. From observations on more than 50 oviducts from adults of different ages, it can be concluded that the visceral muscles of the isolated oviducts showed stable and well-organized contraction patterns when they were taken from a female with an age between 10 and 40 days after adult emergence, when they were submerged in locust saline with a pH ranging from 6.5 to 7.0 and when they were left to stabilize in this saline for minimum 30 min. Removal of trifluoroacetic acid (TFA) out of HPLC purified tissue extracts was very important; even very low concentrations of this compound interfered strongly with the oviduct contraction. Within a certain contraction range, both bovine serum albumin (BSA) and casein could increase the oviduct contraction in a dose dependent manner, suggesting that some non-specific action sites may be present in the visceral muscles of the oviduct.
