Two PVY Spanish isolates from pepper were classified as PVY-1 and PVY-1-2 pathotypes. PVY-1-2 pathotype is reported for the first time in Spain and in the Mediterranean area. These isolates, along with other existing in Spain and belonging to PVY-0 and PVY-1 pathotypes, showed similar serological reaction by immunodiffusion and ELISA tests. Spanish PVY pathotypes inoculated on cultivars other than the usual differential pepper varieties showed differences compared to the corresponding ones in France and Australia. Pepper varieties 'Yolo Y', 'Maor' and 'Italian El' were resistant to pathotype PVY-0; 'Perennial','Florida VR2', 'Delray Bell', 'Ikeda', 'Agronomico-8', 'Agronomico-10' and 'Moura' to PVY-1 and 'Serrano Veracruz', 'Serrano Criollo de Morelos-334', 'Puerto Rico Wonder' and 'LP1' to PVY-1-2. Allelism tests showed that PVY-0 resistance in 'Italian El' is monogenic recessive and its resistance genes allelic or the same to those of 'Yolo Y' (vy1vy1), which resulted also allelic to PVY-0 resistance genes of 'Florida VR2'; the PVY-1 resistance genes of 'Florida VR2', 'Ikeda' and Agronomico-10' resulted to be recessive and allelic; the 'Puerto Rico Wonder' PVY-1 resistance gene was shown to be allelic with vy2 gene of 'Florida VR2'. The resistance of 'Puerto Rico Wonder' to three PVY pathotypes is also cytoplasmically inherited, but the full mechanism must be cyto-genetically operated. Vein necrosis symptoms not explained by monogenic recessive theory were found in some F1s and also a cytoplasmic influence on necrosis expression. Dominant resistance to PVY-1-2 was confirmed in 'Serrano Criollo de Morelos-334' and four kinds of symptoms were checked, but the number of genes controlling them remains to be studied.
