The present paper was conducted by applying paraffin and refrigerated sectioning techniques as well as SEM. The results showed that the differentiation and enation of epidermal cells on the outer integument of cotton ovule have some distinct morphological changes as nucleus enlargement, nucleolus accumulation and increment, vacuole formation and nucleus transferring outwardly. Among these morphological changes, the nucleus and nucleolus may be as the morphological indices of differentiation and enation of fiber. The enation of epidermal cells on the cotton ovules may present phasic characteristics and it may stop between first and second enations in several days.
