Effects of glucose, adonitol or phosphatidylcholine to cooling on in vitro matured bovine oocytes.

The aim of present study was to examine the effects of supplementation of glucose (G), adonitol (AD) or phosphatidylcholine (PC) to the basal 1,2-propanediol (PG) solutions on efficiencies of fertilization, cleavage and development to blastocysts of in vitro matured bovine oocytes exposed at -7 degrees C for 10 min. The following five solutions were used for the present study, 2M PG (2M PG group), 1.5M PG (1.5M PG group), 3) 1.5M PG + 0.5M G (G group), 4) 1.5M PG + 0.5M AD (AD group), 5) 1.5M PG + 0.05% PC )PC group). Firstly, in order to examine the toxicity of the solutions, oocytes were exposed to the respective solutions at 37 degrees C for 10 min. The cleavage and development rate in 2M PG group (p<0.01) and development rate in AD group (p<0.05) were significantly lower than the other experimental groups and untreated group. Secondly, oocytes were exposed to the solutions at -7 degrees C for 10 min. The fertilization rate in 2M PG (p<0.01) and PC groups (p<0.05) significantly decreased compared to untreated group. No significant difference was observed between oocytes exposed at -7 degrees C and untreated controls in polyspermic rate. The cleavage and development rates of oocytes exposed at -7 degrees C were significantly lower than untreated control oocytes. The development rate in G group significantly increased compared to 2M and 1.5M PG groups, while there were no favorable effects of addition of AD and PC on development rate. The result of present study suggested that glucose was effective for protecting in vitro matured bovine oocytes from chilling injury.
