Vaginal irrigation and swab methods were compared in 30 sexually mature bitches with an unknown stage of the estrous cycle, in order to determine if the smears obtained by each method produced a minimal of one hundred differentiable epithelial cells from the vagina and the percentage of cellular damage produced for each method. In addition, to simplify the staining procedure, the Giemsa stain was compared with the Papanicolaou's stain (used as a standard reference through the mature and superficial cell indexes) in order to establish the reliability of the first method to identify and to differentiate vaginal cells. It was established that both methods allowed to get more than one hundred of useful epithelial cells, but the swab method allowed a greater cell number and cellular density. Although this method produced a greater cellular damage than vaginal irrigation, as shown by the exaggerated cellular folding that produced the swab. In addition the sampling procedure and the frottis realization was easier using the swab. There were no differences (p<=0.01) between both stains in terms of cellular identification and differentiation of vaginal cells. Although the Giemsa stain only allowed the analyses by cellular morphology, the cellular percentages obtained were similar to the results obtained using the Papanicolaou's stain. However, this stain has the advantage of staining the cellular cytoplasm according to the cell maturation grade.
