Caratterizzazione di inibitori tripsinici in semi di diverse specie di Medicago.

English. Fourteen accessions of different species of the genus Medicago showing high levels of resistance to insect pests and environmental stresses were characterized for the occurrence of trypsin inhibitory activity (TIA) in the seed. Wide variation was found for TIA among cultivars, the highest values being shown by the annual medics and shrub types, and the lowest content by M. sativa ssp. falcata from the M. sativa complex. A good degree of stability to heat denaturation was also shown by the seed extract of all cultivars tested, with the highest values (100% recovery of TIA after 5 min treatment at 100 deg C) for the M. sativa ssp. x varia and M. sativa ssp. falcata types, and the lowest values (50% recovery) for M. lupulina. The electrophoretic analysis of trypsin inhibitor under native conditions, showed a single protein band, although with different mobilities in eleven populations, whereas multiple bands were present in M. sativa ssp. falcata and M. lupulina strains. The molecular mass of trypsin inhibitor as determined by SDS-PAGE was found to be similar to that of the Bowman-Birk soybean inhibitor. Phylogenetic relationships among the different taxa were inferred by comparing TIA data.

ITALIANO: Quattordici accessioni di specie diverse di Medicago che mostrano elevati livelli di resistenza ai parassiti e agli stress ambientali erano caratterizzati dalla presenza di attivita' inibitoria tripsinica (TIA) nei semi. Si e' riscontrata un'ampia varieta' per TIA nelle cultivar, essendo i valori piu' elevati mostrati dai tipi annuali e arbustivi, e quelli piu' bassi da M. sativa ssp. falcata dal complesso M. sativa. Si e' anche mostrato un buon grado di stabilita' alla denaturazione da calore da parte dell'estratto di semi di tutte le cultivar testate, con i valori piu' elevati (recupero del 100% di TIA dopo un trattamento di 5 minuti a 100 gradi C) per i tipi M. sativa ssp. x varia e M. sativa ssp. falcata, e quelli piu' bassi (recupero del 50%) per M. lupulina. L'analisi elettroforetica dell'inibitore tripsinico in condizioni naturali ha mostrato una singola banda proteica, sebbene con mobilita' diverse in undici popolazioni, mentre bande multiple erano presenti in linee di M. sativa ssp. falcata e M. lupulina. Si e' riscontrato che la massa molecolare dell'inibitore tripsinico determinata in base a SDS-PAGE e' simile a quella dell'inibitore di semi di soia Bowman-Birk. Si sono ipotizzate relazioni filogenetiche tra i diversi taxa attraverso il confronto tra i dati TIA.
