The aim of the study was to analyse the polymorphism of maize lines (Zea mays L.) using sequences of microsatellite DNA and PCR-ISSR markers. Two types of primers have been used. Trinucleotide unanchored primer (ATG)6 and dinucleotide primer AGT(GT)6 anchored at 5 end. Both primers have shown to be effective in order to distinguish a common genetic background of tested lines. Cluster analysis grouped lines having similar genetic background. The line T10 of unknown origin has been separated from the others tested lines at the value of genetic distance 0.9 by using primer (ATG)6. Meanwhile, by using the anchored primer the T10 line has been attached to the lines X4 (Iowa Stiff Stalk Synthetic) and B8 (Iodent Reid x Lancaster Sure Crop x Iowa Stiff Stalk Synthetic) at the value of genetic distance 0.5. Because of higher specificity of anchored primer it may suggest certain similarity of genetic background among those three maize lines. The polymorphism value ranged from 64% (unanchored primer) to 78% (anchored primer).
