文蛤抗癌活性多肽的生理活性研究 | Studies on the Physiological Activity of a Natural Peptide from Clam (Meretrix meretrix Linnaeus )

以新鲜文蛤为材料,经有机溶剂萃取,分子筛凝胶层析,得到一种具有抗癌生物活性的文蛤多肽,SDS PAGE电泳测得该多肽的分子量约为18kD.用该多肽处理人胃腺癌细胞BGC 823,结果表明,当多肽浓度为10μg/mL,细胞生长的抑制率达到50%.此外,探讨了该活性多肽在体外对几种与肿瘤代谢相关的酶的效应,实验结果显示,该活性多肽能够激活碱性磷酸酶和超氧化物歧化酶,当其浓度为10μg/mL时,可以使这两种酶活力分别提高150%和93%;该活性多肽对酪氨酸酶则是起到了抑制的效应,测定导致酶活力下降50%所需的效应物浓度(IC50)为1.50μg/mL,其抑制类型为混合型抑制,其抑制常数KI和KIS分别为4.37mg/mL和6.09mg/mL. 【英文摘要】 A kind of peptide (M_2) was purified from greenness clam (Meretrix meretrix Linnaeus) by the method of alcoholic extraction, gel filtration and so on. The final preparation was homogeneous on SDS-PAGE. The molecular weight was determined to be 18.4 kD by the method of SDS-PAGE electrophoresis. According to the research, this kind of peptide has the inhibitory effect on the growth of human gastric cancer cells BGC-823.
