This investigation was carried out during the period from 2006 till 2010 at Ornamental Horticulture Department, Fac. of Agric., Cairo Univ. Giza. Egypt. The practical Center, Giza. Egypt. This study aimed to: Establish a protocol for Echinacea purpurea plants micro- propagation started by determine the best method of seeds sterilization then culture them to obtain sterilized seedlings .Then evaluate the type of explants and plant growth regulator (PGR) to enhance shoot formation when cultured on MS media containing one kind or a combination of two or three kinds of PGRs. Followed by investigation of two kinds of auxins on root development, then investigation the proper medium for plant adaptation. The most efficient period for seeds by using H2O2 (30% v/v). The best germination percentage was (38.89%) registered with coated seed sterilization was 15 min. for decoated seeds and 30 min. for coated seeds by using H2O2 ( 30% v/v) .The best germination percentage was ( 38.89%) registered with coated seeds sterilized by ( 30% v/v) H2O2 and incubated in light K PGR produced the highest mean number of shoots (17.33 shoots/jar ) which produced directly from leaf explants at 2mg/L. Combination of BA+NAA+K at concentrations 0.5+0.1+1.0 mg/L resulted in the highest mean number of shoots (24 shoots/jar) from root explants. Addition of IAA at 0.5mg/L to culture medium gave the highest mean number of formed root (5.66 roots/shoot ) Using medium consisting of sand : peatmoss 1:1 (v/v) was the most favorable medium for plant acclimatization. Study the effect of different types and concentrations of auxins on anthocyanins pigments accumulation in calli cultures which produced from leaf and root explants. The highest value (0.122 mg/l00gm root) was obtained from red roots which formed naturally in vitro, the highest value 0.089mg/100gm of root callus had been obtained on media supplemented with (1.0mg/L NAA + 3 .00 mg/L K) produced of callus as the highest value of treatments. Investigate the effect of some kinds of precursors (caffeic acid _caffeine/ tryptophan) at the presence of NAA to enhance the accumulation of active ingredients in root callus on solid media. The highest value 0.055mg/gm callus was obtained from cullus formed on medium supplemented by (1mg /L NAA + 15 mg/L CA). Compare the content and concentrations of active ingredients between roots produced in vitro and precursor's treatments by using HPLC. Red root produced the highest total amount of flavonoides (23.3 mg7gm root) and the total amount of phenoilc compounds (300.20 mg/gm root).
