[The Effect of Insecticides on Insect Cell Culture in Vitro]

The cell line Sf9 derived from a pupal ovarian tissue of Spodoptera frugiperda was cultured, subcultured(23 subculture) and cryopreserved. Morphological characterization of Sf9 cells was performed through using three staining procedure ( Trypan blue, Wright, Giemsa). The Sf9 cell population demonstrated distinct cells with three different shapes : spherical cells (large one and small one); fibroblast-like cells, and Phagocytic cells, Also, Two different populations of cells were identified in Sf9 cell cultures according to nuclear number: mononucleated and polynucleated . In this study three insecticides :the organophosphate insecticide 'Dimethoate', the Neonicotinoid insecticide' Acetamiprid' and the Pyrethroid insecticide 'Deltamethrin' that have according to references different effects and modes of action were evaluated in vitro on cultured Sf9 cell line concentration range (0.5 to 500 μM). The cell growth inhibition was measured by tetrazolium (MTT) assay. Analysis of regression was used to estimate the 50% and 20% inhibition cell growth (IC50, IC20). Toxicity bioassays indicated that Deltamethrin was the most toxic one (IC50=132.5 μM, IC20=46.8 μM). The number of Phagocytic cells was positively correlated with the concentration of insecticide in culture media. This research presents an important example for the useful application of in vitro insect cell culture in the prescreening of insecticide's effect ; especially, newly introduced ones.
