Actinomycetes from rice field soil and their activities to inhibit rice fungal pathogens

Total of 247 actinomycetes were isolated from the 35 rice-field soil samples collected from 16 provinces in Thailand. The studied of morphological characteristics and types of diaminopimelic acid in whole cell hydrolysates revealed that 198 isolates were classified into the group of streptomycetes, with the remaining 49 isolates were classified into non-streptomycetes. All isolates were tested for their ability to inhibit the growth of three rice fungal pathogens: Fusarium moniliforme, Helminthosporium oryzae and Rhizoctonia solani on potato dextrose agar by dual culture technique. Isolate RF 16-12 which showed the highest antifungal activities against all tested fungi was determined the effect on the germination of rice seed in the presence and absence of F. moniliforme using sterilized moisture paper method. Data analysis by SPSS using Least Significant Difference (LSD) test showed that spore suspension of RF 16-12 was effective in controlling F. moniliforme on rice seed with no effect on root and shoot development of rice seed. Molecular identification by sequencing of 16S rRNA gene revealed that the isolate RF 16-12 shared 99.86 percent similarity with Streptomyces yogyakartensis NBRC 100779 sup(T).
