In vitro response of some Potato cultivars to salt stress

In this study, The in vitro responses of different potato cultivars (cvs) to salt stress (0.0, 1.0, 2.0, 3.0 and 4.0 g/L) were investigated based on changes in shoot and root growth (first experiment) and microtuberization potential (second and third experiments). Our results indicated that increasing salt concentration in the tissue culture medium generally decreased plantlet fresh weight (FW), plantlet height, number of nodes per plantlet and root length. Shoot and root lengths were more affected by salt stress than nodes number and FW. Differences among ten potato cultivars in response to salinity were observed. Based on the percentage of shoot height value at the highest NaC1 level (4.0 g/L) relative to the control, potato cvs showed the following variable pattern of salinity tolerance: ArindaDiamantSante BolistaSilvesterArnova Atlas Proventa Picasso Spunta. Microtuber yield /Jar and average microtuber weight increased at NaCl level ranges from 1.0 to 3.0 g/L ,and was similar to the control at 4.0 g/L NaCI. Number of microtuber per jar and percent tuberization from single node explants increased at 1.0-2.0 g/L NaCI, then declined at 4.0 g/L. Based on their microtuberization potential, the two cvs f Agria and Arinda were ranked as the most tolerant, and the cv Safran as the most sensitive, while the cvs Marabel, Bolista, Sante, Proventa, Universa, Picasso and Spunta were moderately tolerants. Salt stress decereased chlorophyll and .J carotenoid contents, whereas increased accumulation of proline, super oxide dismutase and catalase activities in potato tissue, depending on the cultivar.
