Optimization of hyperimmunization process in the production of snake antivenin serum

One small-scale double emulsion technic for incorporation of Naja- Naja oxiana venom Poly (lactide-co-glycolide) (PLG) microspheres and bentonite adsorbed suspension were developed and optimised. The effects of high speed homogenisation on the double emulsion, stability, microsphere size, entrapment efficiency and In vitro release of venom were studied. A stable double emulsion was verfied by homogenisation method. Slow removal of the organic allowed measurment of the size of the emulsion droplets and subsequent predication of the size of the resulting micropheres, Microspheres in the size range of 1-1O µm were prepared using homogenisation technic, but this tech, was sensitive to changes in the operating time, speed and volume of outer aqueous phase. Snake venom was released in vitro in a triphasic manner. After immunisation of guinea - Pig with a two 1M injection, the PlG - venom microspheres, an antibody response very high as that elicited wlth conventional method and bentonite adsorbed suspension and venom alone, while bentonite system elicited response equal to conventional system. these results indicate that the antigenicity of venom was retained after oration into PLG microspheres using homogenisation technic, and adsorption on bentonite, but microphere have higheradjuvant effect.
