Simple and rapid colorimetric method for phytate analysis in rice

Rice is the primary food source for more than half of the world population. In the Philippines, it is the major staple food and, as such, is an important source of essential human nutrients, including carbohydrates and some vitamins and minerals. Zinc is one of the essential micronutrient minerals in humans, but zinc deficiency has been identified as a significant issue in human nutrition in the Philippines. Because of the large amount consumed per person, rice has the potential to become an important source of dietary zinc. However, as with many plant sources, rice also contains significant amounts of the anti-nutrient phytate (inositol hexakisphosphate (IP6)). It is a storage form of phosphate that is used by the seed during germination. It has been shown that phytate interferes in the absorption and utilization of divalent minerals such as iron, copper, calcium, and zinc, thus making the minerals less available to human consumers. Reports showed that zinc, particularly during its absorption in the digestive tract and its pancreatic recycling, remains the most vulnerable of the essential cations complexed with phytate, which can lead to zinc deficiency. When assessing the possibility of improving the Zn content of rice, it is of considerable interest to study the phytate: zinc molar ratio in the grains to evaluate available zinc for human nutrition. The objective of this study is to modify the colorimetric methods available for phytate analysis to design and validate a more accurate, simpler, and less expensive colorimetric technique for assaying a large number of rice samples, which can be applied in breeding and screening of large number of rice varieties.
