Isolation and characterization of Bacillus species antagonistic to soil-borne fungal pathogens

The genus Bacillus is a heterogeneous group of endospore-forming bacteria isolated from the soil. These microorganisms have been used successfully as biocontrol agents against many soil-borne fungal pathogens. This study was conducted to isolate Bacillus strains capable of inhibiting the growth of Colletotrichum falcatum, Fusarium oxysporum, Macrophomina phaseolina and Alternaria alternata and also to analyze the diversity of Bacillus in different rhizospheric soil samples. Soil samples were collected from different areas of Pakistan. Out of fifty isolates ten exhibited efficient antagonistic activity against these four fungi. Bacillus mycoides strain S1 showed maximum inhibition of C. falcatum followed by Brevibacterium strain LB44 and Bacillus pumilus strain S5. Enterobacter strain LB41 showed maximum antimicrobial activity against A. alternata followed by Brevibacterium strain LB44 and Bacillus subtilis strain S6. Bacillus pumilus strain ME42 showed maximum inhibition of M. phaseolina followed by Enterobacter strain LB41 and Brevibacterium strain LB 44. Bacillus pumilus strain S5 showed antimicrobial activity against F. oxysporum followed by Bacillus subtilis strain S6 and Bacillus mycoides strain S1. Further biochemical and morphological studies of these potential strains were done which suggested that these strains produced siderophores and other secondary metabolites as hydrolyzing enzymes which could be effective against different fungi.
