A relationship between ploidy level and the number of chloroplasts in stomatal guard cells in diploid and amphidiploid Brassica species | Связь плоидности с числом хлоропластов в замыкающих клетках устьиц у диплоидных и амфидиплоидных видов Brassica

During production of doubled haploid lines through isolated anthers and isolated microspores cultures the ploidy level of microspore derived plants varies, and normally haploid, diploid and mixoploid plants could be obtained in vitro. The determination of ploidy level by counting the number of chloroplast in stomatal guard cells (NCSGC) is the least time consuming, laborious and expensive. The NCSGCs of haploid (n) and diploid (2n) Chinese cabbage (Brassica rapa ssp. pekinensis), winter oilseed rape (B. napus var. napus) and haploid, di- and tetraploid white cabbage (B. oleracea var. capitata) microspore derived plants were estimated, and also the influence of plant growth temperature (6±2 deg C and 24±2 deg C) and development stage (vegetative or generative) was investigated. High correlation between the ploidy level of microspore-derived plants and NCSGC is found for white cabbage (r = 0.94), Chinese cabbage (r = 0.90) and oilseed rape (r = 0.94). The average NCSGC was very similar among the same ploidy genotypes of Chinese cabbage as well as rapeseed, while the variation of chloroplast number in diploid and tetraploid white cabbage plants was significant. In a range of early, middle and late maturing diploid white cabbage inbred lines there was established the tendency to form more chloroplasts in the early lines (Pl, Sus and others) and less in the late lines (AM2, Sa1, Ges2 and others), with the difference up to 1.7 times, that is comparable to the difference between haploid and diploid plants of Chinese cabbage or rapeseed. The chloroplast number in stomatal guard cells is 4.2-7.8 and 7.9-13.6 for Chinese cabbage haploids and diploids, respectively, 7.5-12.4 and 14.1-20.3 for rapeseed amphihaploids and amphidiploid, respectively, and 7.7-9.9, 11.7-17.9 and 18.0-26.5 for white cabbage haploids, diploids and tetraploids, respectively. No significant influence of vegetative or generative stage of plant development or growth temperature on NCSGC.

Russian. При создании удвоенных гаплоидов в культурах пыльников и изолированных микроспор среди получаемых in vitro растений часто встречаются гаплоиды, тетраплоиды и миксоплоиды. Определение плоидности по числу хлоропластов в замыкающих клетках устьиц (ЧХЗКУ) отличается простотой и низкой стоимостью анализа. У ряда представителей рода Brassica известно ЧХЗКУ, характерное для разных уровней плоидности. Проведен сравнительный анализ ЧХЗКУ для листьев гаплоидных и диплоидных растений капусты пекинской B. rapa ssp. pekinensis (КП), озимого рапса B. napus, гаплоидных, ди- и тетраплоидных растений капусты белокочанной B. oleracea (КБ). Изучено влияние на проявление признака температурных условий (6±2 град. С и 24±2 град. С), стадии развития (вегетативная - 4-5 настоящих листьев, генеративная - цветение) и генотипа. В исследовании использовали растения-регенеранты, полученные в культуре изолированных микроспор, инбредные линии, коммерческие сорта и гибриды F1. Установлено, что ЧХЗКУ не относится к генотипспецифичным признакам у КП и рапса. У инбредных линий КБвыявлена зависимость ЧХЗКУ от скороспелости и показана тенденция к формированию большего ЧХЗКУ у раннеспелых линий (Пл, Сюс и др.) и меньшего - у позднеспелых (AM2, Са1, Гэс2 и др.). При этом у диплоидных скороспелых инбредных линий ЧХЗКУ оказалось в 1,7 раза больше, чем у позднеспелых, что сопоставимо с различиями между гаплоидными и диплоидными растениями КПили рапса. Среднее ЧХЗКУ у гаплоидных растений КП c составило 4,2-7,8, у диплоидных - 7,9-13,6; у амфигаплоидных растений рапса - 7,5-12,4, амфидиплоидных - 14,1-20,3 шт.; у гаплоидных растений КБ- 7,7-9,9, диплоидных - 11,7-17,9 и тетраплоидных - 18,0-26,5 шт. Выявлены высокие коэффициенты корреляции между плоидностью и ЧХЗКУ у растений КБ, КП и рапса (соответственно r = 0,94, r = 0,90, r = 0,94). Признак ЧХЗКУ не изменяется при переходе от вегетативной к генеративной стадии развития и не подвержен влиянию температурного фактора.
